Reversal of pyruvate oxidation in Clostridium butyricum.

چکیده
The failure to detect an equilibrium between acetyl phosphate, a product of pyruvate cleavage, and pyruvate has been one of the poorly understood observations of the pyruvate cleavage reactions in Clostridium butyricum and in Escherichia coli (l-3), although pyruvate synthesis from acetyl phosphate was reported in the latter organism (4). In the preceding paper (5) a small incorporation of acetyl-lCY-CoA into pyruvate was observed with extracts of C. butyricum. Since these extracts rltpidly equilibrate CO?. with the carboxyl group of pyruvate (3), the poor reversibility of the elastic system appears to lie in the nature of the oxidative portion of the reaction. Peck and Gest (6) have found that the electron transport system in these extracts may be activated by sodium hydrosulfite. The following report deals with pyruvate formation from acetyl phosphate and COZ in extracts of C. butyricum and the stimulation of this reaction by sodium hydrosulfitc.

Cofactors of the phosphoroclastic reaction of Clostridium butyricum.
The degradation of pyruvate, in the presence of phosphate, to acetate, carbon dioxide, and hydrogen by cell-free extracts of Clostridium butylicum was described by Koepsell and Johnson (1). Later acetyl phosphate, the labile precursor of the acetate, was shown to be produced (2, 3), and phosphotransacetylase was shown to be present in the cell-free extracts (4). This reaction is, at least, supe...
متن کاملCarbon dioxide activation in the pyruvate clastic system of Clostridium butyricum.
Extracts of Clostriclium butyricum catalyze a rapid exchange reaction between COZ and the carboxyl group of pyruvate (1, 2). This reaction has been reported to require diphosphothiamin, coenzyme A, and phosphate (or arsenate) (2). These cofactor requirements are essentially the same as those required for the degrada.tion of pyruvate to acetyl phosphate, Con, and Hz by this organism (3). The com...
متن کاملPyruvate Exchange Reactions in Bacillus Macerans1
The fermentation of carbohydrates by Bacillus macerans yields ethanol, acetone, acetic and formic acids, hydrogen, and carbon dioxide (Schardinger, 1905; Donker, 1926). In bacteria, hydrogen is known to arise anaerobically through pyruvate cleavage which also yields acetate and CO2. The most thoroughly studied of these reactions are the phosphate-requiring cleavage reactions in Escherichia coli...
متن کاملMolecular characterization of the 1,3-propanediol (1,3-PD) operon of Clostridium butyricum.
The genes encoding the 1,3-propanediol (1,3-PD) operon of Clostridium butyricum VPI1718 were characterized from a molecular and a biochemical point of view. This operon is composed of three genes, dhaB1, dhaB2, and dhaT. When grown in a vitamin B12-free mineral medium with glycerol as carbon source, Escherichia coli expressing dhaB1, dhaB2, and dhaT produces 1,3-PD and high glycerol dehydratase...
متن کاملComparative aspects of fatty acid activation in Escherichia coli and Clostridium butyricum.
The role of acyl-ACP in the acylation of glycerol3-phosphate has been recently demonstrated in E. coli and C. butyricum [l-3]. In E. coli, both CoA and ACP derivatives can serve as substrates for this reaction [4], whereas in C’. butyricum ACP derivatives are used almost exclusively; the biosynthesis of the latter compounds is known to proceed only through the fatty acid cycle [5] . The possibl...
